OCR: A restriction map is a physical map of a cloned DNA segment that shows the positions of cutting sites for different restriction enzymes. In the illustration, an 8-kb DNA molecule is digested in separate reactions with the restriction enzymes EcoRI and BamHI. The resulting fragments are then separated according to size by gel electrophoresis, and their lengths are determined by comparison with known standards. As illustrated, either of two possible locations of the BamHI site relative to the EcoRI site are consistent with the electro phoresis results. A third digestion with both enzymes reveals the correct map.
